Synonyms: CMS 121, Fisetin derivative, Salk CMS-121
Description:
CMS-121 is a fisetin-derived neuroprotective compound developed for its potent effects on cognitive preservation, mitochondrial health, and metabolic regulation. Engineered to improve bioavailability and blood-brain barrier permeability over fisetin, CMS-121 has demonstrated robust anti-aging and neuroprotective benefits in preclinical studies. It is currently under investigation as a geroprotective agent in research on Alzheimer’s disease, cognitive decline, and oxidative stress reduction.
Mechanism of Action:
- Activates AMPK and PGC-1α, enhancing mitochondrial biogenesis
- Reduces lipid peroxidation and oxidative stress
- Protects neurons against toxic protein accumulation
- Improves synaptic plasticity and brain energy metabolism
Key Research Areas:
- Alzheimer’s and dementia models
- Age-related memory decline
- Mitochondrial dysfunction and neuroinflammation
- Cognitive resilience and lifespan extension
History of Discovery:
CMS-121 was developed by The Salk Institute for Biological Studies as a next-generation derivative of fisetin, a natural flavonoid with known neuroprotective effects. By structurally modifying fisetin to increase stability and brain penetration, CMS-121 has emerged as one of the most promising compounds in the Cellular Geroscience field.
Case Studies:
- Alzheimer’s Disease Model (2019, eLife): CMS-121 preserved cognitive performance and reduced oxidative brain damage in transgenic Alzheimer’s mice. [eLife. 2019;8:e46994.]
- Mitochondrial Function in Aging Brains (2020, Aging Cell): Improved mitochondrial respiration and reduced lipid peroxidation were observed in aged mice treated with CMS-121. [Aging Cell. 2020;19(2):e13021.]
- Metabolic Enhancement in Senescence Models (2022, J Gerontol A Biol Sci Med Sci): CMS-121 activated AMPK and improved insulin sensitivity in aging rodents. [J Gerontol A Biol Sci Med Sci. 2022;77(5):899–907.]
