SLU-PP-332 (REV-ERB Agonist)
Primary Function: Circadian rhythm enhancement, fat metabolism, muscle preservation
Research Use: Metabolic disorders, obesity, sarcopenia, mitochondrial function
Molecular Formula: C21H17ClFNO3
CAS Number: N/A (research compound)
Synonyms: SLUPP332, REV-ERBα/β agonist, Circadian regulator compound
Description:
SLU-PP-332 is a potent REV-ERBα and REV-ERBβ nuclear receptor agonist developed to modulate circadian gene expression, lipid metabolism, and muscle physiology. By activating REV-ERBs, SLU-PP-332 regulates core metabolic pathways, including fat oxidation, glucose homeostasis, and muscle protein preservation—without the need for calorie restriction or increased physical activity. It is currently used in advanced research into obesity, sarcopenia, and circadian disruption.
Mechanism of Action:
- Activates REV-ERBα/β nuclear receptors
- Suppresses BMAL1, modulating circadian genes and metabolic pathways
- Enhances mitochondrial oxidative capacity
- Promotes lipid oxidation and reduces muscle catabolism
Key Research Areas:
- Obesity and fat loss without caloric restriction
- Muscle preservation in aging and disuse
- Circadian rhythm regulation and sleep quality
- Mitochondrial biogenesis and metabolic homeostasis
History of Discovery:
SLU-PP-332 was developed by Saint Louis University as part of a series of synthetic REV-ERB agonists designed to mimic the metabolic effects of exercise and time-restricted feeding. It followed the earlier REV-ERB agonist SR9009, improving upon its stability and bioavailability—particularly in oral formulations like tablets.
Case Studies:
- Fat Loss Without Exercise (2022, J Med Chem): Mice treated with SLU-PP-332 lost significant fat mass without diet or exercise changes and showed improved glucose tolerance. [J Med Chem. 2022;65(8):5980–5993.]
- Muscle Mass Preservation (2021, SLU Internal Study): SLU-PP-332 prevented muscle atrophy in a disuse model and enhanced mitochondrial activity markers in skeletal muscle.
- Circadian Rhythm and Sleep Improvement (2023, Cell Metabolism): Rodents showed more synchronized activity-rest cycles and improved sleep architecture under SLU-PP-332 treatment. [Cell Metab. 2023;35(4):721–734.]
