Melanotan 2 (MT-2)

Primary Function: Tanning peptide, libido enhancement, photoprotection
Research Use: Skin pigmentation, UV protection, sexual function, metabolic modulation
Molecular Formula: C50H69N15O9
CAS Number: 121062-08-6
Synonyms: MT-2, Melanotan II, Melanotan-2, MTII

Description:
Melanotan 2 is a synthetic analog of α-MSH (alpha-melanocyte-stimulating hormone) that binds to melanocortin receptors, particularly MC1R and MC4R. It is extensively studied for its ability to stimulate melanin production, resulting in skin darkening without prolonged UV exposure. Additionally, MT-2 has shown effects on libido enhancement, appetite suppression, and energy balance, making it a popular subject in cosmetic, metabolic, and sexual health research.

Mechanism of Action:

  • Stimulates melanogenesis by activating melanocortin 1 receptors (MC1R)
  • Influences libido and erectile response through melanocortin 4 receptor (MC4R)
  • Enhances natural UV defense mechanisms via increased melanin production
  • May affect appetite, mood, and energy regulation

Key Research Areas:

  • Sunless tanning and skin pigmentation
  • Libido and erectile function enhancement
  • Photoprotection and UV-related skin health
  • Energy metabolism and appetite control

History of Discovery:
Melanotan 2 was developed at the University of Arizona in the 1990s as a sunless tanning agent to reduce UV exposure and skin cancer risk. During trials, unexpected libido-enhancing effects were observed, which sparked additional studies into its influence on sexual function. It has since become one of the most researched peptides in cosmetic and sexual wellness applications.

Case Studies:

  1. Tanning and UV Protection (2000, J Photochem Photobiol B): MT-2 significantly increased skin pigmentation in fair-skinned subjects with reduced UV exposure. [J Photochem Photobiol B. 2000;56(1):106–113.]
  2. Erectile Function Improvement (2004, J Urol): MT-2 enhanced erectile response in men with psychogenic ED, even without sexual stimulation. [J Urol. 2004;171(6 Pt 1):2713–2716.]
  3. Reduced Sunburn Risk (2009, Arch Dermatol Res): MT-2 treatment resulted in lower rates of sunburn and DNA damage after UV exposure. [Arch Dermatol Res. 2009;301(6):439–444.]
